唐朝贞观九年利州都督武士镬得了一名千金取名武媚。14岁时的武媚娘被召入宫中服侍太宗更衣，与淡泊名利的九皇子晋王李治日久生情。太子被废后，媚娘揭发四皇子李泰罪行使得李治当上太子顺利继承帝位，可是媚娘却不得不奉太宗遗诏，削发为尼进入感业寺修行，幸得智空和尚相助得以回宫。回宫后的媚娘与王皇后勾心斗角，使出浑身解数的媚娘终于被立为皇后。武后为竭尽心力辅佐朝政，招来旧相识程南英与明学文为智囊，又请来胞姐贺兰夫人抚养孩子。李治身体日渐衰竭，与皇帝同时听政的武后渐渐忽略了丈夫和儿子，变得专横独断。在经历了儿子李贤谋反、废帝重立、徐敬业揭竿起义之后，武后终于自立为皇帝，同时也开始了众叛亲离的后半生
