巨大な組織力と機械怪物で構成され、首領のアイアンクロー（鉄の爪）に率いられる犯罪シンジケートクライムの暗躍に際し、国際科学特捜隊日本支部長官の鯨井大助（ジョーカー）に招集された桜井五郎・東竜・カレン水木・大地文太の4人は、核・電気・磁力・重力のエネルギーを持つサイボーグ戦士ジャッカーとして生まれ変わり、トランプのコードネームを持つジャッカー電撃隊となる。ジャッカーはクライムの陰惨な組織犯罪を捜査して壊滅を目指すが、宇宙的科学力にスケールが拡大していくその攻勢に苦戦を強いられる。そんな中、行動隊長の番場壮吉（ビッグワン）が加わり、5人になったジャッカーは、クライムと雌雄を決するべく戦いを繰り広げていく。
